Abstract
AIM: To observe the insulin sensitizing effect and insulin resistance ameliorating effect of rosiglitazone. METHODS: On the IDDM model prepared by the intravenous injection of streptozotocin (50 mg · kg-1), the decrease of blood glucose of only rosiglitazone, only a low dose of insulin (2.5 U · rat-1, sc), rosiglitazone plus a low dose of insulin on the models were calculated; On the immune model of insulin resistance prepared by the intravenous injection of bacillus calmette guerin (BCG, 10 mg · kg-1), the increase of glucose infusion rate (GIR) of rosiglitazone on the model was measured. RESULTS: At three doses of 1, 3, 10 μmol · kg-1 (po on successive 8 days), rosiglitazone decreased the blood glucose values neither of the normal rats nor of the diabetes rats induced by streptozotocin. However, rosiglitazone, at the doses of 3, 10 μmol · kg-1 (po on successive 8 days) plus a dose of insulin (2.5 U · rat-1 · d-1, sc on successive 8 days), decreased the blood glucose values of the diabetes rats induced by streptozotocin. Rosiglitazone, at a dose of 10 μmol · kg-1 (po on successive 8 days) increased the GIR (glucose infusion rate) of the model, determined by both submaximal hyperinsulinemic euglycemic clamp and maximal hyperinsulinemic euglycemic clamp. CONCLUSIONS: Rosiglitazone possesses the insulin sensitizing effect and insulin resistance amelioration effect.
