In vitro RNA synthesis with SP6 RNA polymerase

Abstract

In this chapter we describe the use of in vitro transcription systems for the synthesis of RNAs for use as substrates and hybridization probes. We discuss advantages of the technique and several associated problems. Because our experience is primarily with SP6 RNA polymerase we will use this system as an example throughout, but we believe the observations are general and apply to any of the phage polymerase and promoter systems, including T3 and T7.
