Effect of magnesium sulfate on nitric oxide production in cultured human umbilical vein endothelial cells

OBJECTIVE: Nitric Oxide (NO) is a potent vascular endothelial cellderived relaxing factoi. Magnesium sulfate (MgSO4) has been shown to promote vascular endothelial production ot proslacyclin. We studied the e fleet ot MgSO4 on NO production in cultured human umbilical vein endothe-lial cells (HUVEC). STUDY DESIGN: IU'VEC were incubated uiih different concentrations ol MgSO4 (0, O.n, 3. and 5 niM, n - S fach) for 24 hours. The concentrations of 0.1 inM and 3 mM of MgSO4 are reported to be pinsilogic and theiapeutic concentrations, respectively. After 24 hours of UK libation, the media uere colleclfd lor NO anaivsis. NO was measured by monitoring the toi ma t ion ot total nit lie tide (TNOx: NO + nitrite [NO2-1 + nitrate [NOS-J) l> a chemiluniinescence detector after reduction of TNOx to NO by acidic vanadium III. The alue was quantitaled using an integrator by reference to NaNO, standards. Results were adjusted by the amount of cellular protein in eat h well. Anaivsis of variance was used for statistical anaivsis. Results aie given as means ±SF.M in pmolmg protein. RESULTS: Magnesium sulfate concentrations 0 mM 0.6 mM 3 mM 5 mM (N =8) (N = 8) (N = 8) (N = 8) P Nitric oxide 30.1 ± 2.8 : 32.5 ± 3.6 35.3 ±. 5.4 :W.H ±4.7 NS (pmol/mgn protein) CONCLUSIONS: Ahhough magnesium suliate promotes vascular endothelial cell production ol prostanclin. our data do not show that IgSO4 enhance-, the in vitro production ot NO cultured HLVEC. Il indicates that MgSO4-induced vasodilatation niav not be mediated through (Miham ed nitric oxide pioduclion in IRA'F.K.
