Abstract
Microbial contamination of high-purity water is a significant problem in semiconductor, pharmaceutical and food and beverage industries. Bacterial contamination in different parts of a high-purity water system was studied using plate counts, epifluorescence microscopy, and 16SrRNA gene sequencing. Oligotrophic bacteria strains were classified. Methods requiring direct culture of the bacteria failed to correctly estimate the extent of bacterial contamination. Use of membrane filtration followed by epifluorescence microscopy allowed the rapid and accurate determination of biocontamination levels.
